Ribon Therapeutics
The business, the turning points, and the decisions behind them.
01 / WHAT IT BUILT
What the company did
Ribon Therapeutics was a Cambridge, Massachusetts-based clinical-stage biotech founded in 2015. It developed precision medicines targeting stress-support pathways — specifically first-in-class small molecule inhibitors of NAD+-utilizing enzymes that become pathologic under stress. Its most mature candidate, RBN-2397, was a PARP7 inhibitor being developed for squamous cell lung carcinoma in combination with Merck's Keytruda; a second candidate, RBN-3143, was a PARP14 inhibitor for atopic dermatitis. The company was backed by big pharma investors including AbbVie, Bristol Myers Squibb, Johnson & Johnson, Novartis, Pfizer and Alphabet's GV, raising about $259 million.
02 / Ribon Therapeutics FAILURE REASONS
Why it failed
After nearly a decade in business, Ribon ran out of runway: in May 2024 it announced it had 'discontinued all material operations' and put substantially all of its assets — primarily intellectual property and its clinical development programs — up for online auction, including its interest in deferred and contingent milestone payments owed by Boehringer Ingelheim. Despite an impressive roster of pharma backers and generally safe and well-tolerated Phase I data, the company could not secure a path forward.
