Spotlight Bio\USA
The business, the turning points, and the decisions behind them.
01 / WHAT IT BUILT
What the company did
Spotlight Therapeutics (listed as Spotlight Bio) was a San Francisco biotechnology company developing in vivo CRISPR gene editing therapies. Its proprietary TAGE (Targeted Active Gene Editor) platform used a cell-targeted, non-viral delivery system - structured like an antibody-drug conjugate but carrying a CRISPR-Cas payload - designed to deliver gene editors directly to cells without lipid nanoparticles or viral vectors. Founded in 2018 by scientists Alex Marson, Jacob Corn, and Patrick Hsu and led by CEO Mary Haak-Frendscho, the company raised a $30 million Series A led by GV. Its therapies never reached the clinic; the company shut down in early 2025.
02 / Spotlight Bio\USA FAILURE REASONS
Failure reasons
Spotlight shut down because its delivery platform was not mature enough to work. The TAGE platform required far greater in vivo stability for systemic administration than it could achieve, forcing the company to abandon its original immuno-oncology approach - knocking out ADAR in CD11a-positive immune cells in solid tumors - and pivot to ophthalmology, where local administration into the eye was more feasible. But the preclinical data did not hold up: a preclinical study edited only about 7% of retinal cells when injected directly into mouse eyes, and the rate dropped below 1% in minipigs. With no path to the clinic, the company closed.
03 / EVENT HISTORY
Events in the structured record
- Company founded
Spotlight Therapeutics was founded in San Francisco by Alex Marson, Jacob Corn, and Patrick Hsu to develop in vivo, cell-targeted CRISPR therapies using its TAGE platform.
- $30M Series A
The company closed a $30 million Series A financing led by GV to advance its non-viral gene editing programs in hemoglobinopathies and immuno-oncology.
- Weak preclinical data
A preclinical study published in late 2024 showed the TAGE platform edited only about 7% of retinal cells in mice and below 1% in minipigs, after the company had pivoted from immuno-oncology to ophthalmology.
- Company shuts down
Former CEO Mary Haak-Frendscho confirmed to Fierce Biotech that Spotlight had shut down, after concluding its platform was not sufficiently mature for systemic administration.

05 / CAUSAL CHAIN
How the factors connect
- 01
In early 2025 Spotlight Therapeutics shut down, confirmed by former CEO Mary Haak-Frendscho.
- 02
The company had raised a $30 million Series A led by GV; no public disclosure of total losses, debts, or creditor claims was found.
- 03
No successor, acquirer, or asset sale was publicly reported - the company closed because its TAGE platform could not reach the clinic.
