01 / WHAT IT BUILT

What the company did

Inspirna was a New York City-based cancer biotech company, originally founded as RGENIX from science at Rockefeller University and renamed Inspirna in 2021. It developed first-in-class drugs targeting key pathways in cancer progression using a microRNA-based target discovery platform called RNA-DRIVEr, which identified novel therapeutic targets in the tumor microenvironment. Its lead therapy, RGX-104, was a small-molecule immunotherapy blocking tumor immune evasion that obtained FDA Orphan Drug Designation in several solid tumor types and entered clinical trials in late 2016. The company raised more than $140 million from investors including Novo Holdings, Sofinnova Partners, Exor and Vivo Capital, and wound down in March 2025.

02 / Inspirna FAILURE REASONS

Failure reasons

CEO Usman Azam announced the wind-down on LinkedIn on March 31, 2025, saying the company was closing completely after ten years, but gave no official reason for the decision and no details on staff numbers. The company's last financing was a $50 million Series D in July 2022, roughly three years before the closure; no pipeline or financial catalyst was disclosed, and no postmortem has been published. The closure fits a broader 2025 trend of private biotechs shutting down, but nothing specific to Inspirna's reasons is on the record.
