Spotlight Therapeutics
The business, the turning points, and the decisions behind them.
01 / WHAT IT BUILT
What the company did
Spotlight Therapeutics was a San Francisco-based CRISPR gene-editing biotech founded in 2018 by scientists Alex Marson (UCSF), Jacob Corn (ETH Zurich), and Patrick Hsu (UC Berkeley), led by CEO Mary Haak-Frendscho. It developed the TAGE (Targeted Active Gene Editor) platform - a cell-targeted, non-viral in vivo gene-editing system structured like an antibody-drug conjugate with a CRISPR-Cas payload: a cell-targeting domain, a cell-penetrating peptide, and a Cas protein loaded with guide RNA. The approach aimed to skip lipid nanoparticles and viral vectors and deliver editing directly to desired cell types, initially targeting hemoglobinopathies (sickle cell disease) and immuno-oncology (reprogramming the tumor microenvironment). It raised $30 million in Series A in November 2020 led by GV (formerly Google Ventures), and $66 million total. The company shut down after disappointing preclinical results; the closure was confirmed by the former CEO to Fierce Biotech.
02 / Spotlight Therapeutics FAILURE REASONS
Failure reasons
Spotlight Therapeutics shut down after disappointing preclinical results, the former CEO confirmed. The core problem was the delivery platform itself: the TAGE system was not sufficiently matured to support systemic administration, which requires significantly greater in vivo stability. Spotlight's original immuno-oncology approach - knocking out ADAR in CD11a-positive immune cells in solid tumors - was abandoned as the team pivoted to ophthalmology, where local administration was more feasible. But even there, a preclinical mouse study published on bioRxiv in December 2024 showed the asset edited only about 7% of retinal cells when injected directly into the eye; in minipigs the rate dropped below 1%. With the platform unable to demonstrate the delivery efficiency needed for systemic or even local therapy, investors did not continue funding.
